Reading a certificate of analysis
What each method establishes
| HPLC | Chromatographic purity — the proportion of the sample that is the target peptide rather than related impurities. |

| Mass spectrometry | Identity — that the measured molecular mass matches the expected mass for the sequence. |
A certificate that reports one without the other establishes purity without identity, or identity without purity. Both are needed.
